I'm trying to boot on a USB hard drive and I have a blue screen Stop
0x0000007b.
If I boot the same files from a USB Flash drive, it works...
Why my build cannot boot on USB Hard Drive ?
The drive if formated in NTFS like the Flash drive....

Nick,
Very first thing to check if you have all the components listed here: http://msdn2.microsoft.com/en-us/embedded/aa731218.aspx
If all looks good, try to use debug version of the ntdetect.com: USBNTD.CHK. Read more in the docs:
http://msdn2.microsoft.com/en-us/library/aa940870.aspx

In addition to what KM has provided, there are specific components that you
want to have in the image:

Note: Microsoft has tested the USB Boot solution on USB flash drives, but
they haven't discused testing on USB hard drives.
